Effects of growth factor deprivation on MKN-45 spheroid cells

Background/aim: Serum and growth factor deprivation, a common cellular stressor in solid tumors, arises upon irradiation, chemotherapy, antiangiogenesis. Spheroid body culture aims to enrich cancer stem cells by using low attachment conditions some factors, such as basic fibroblast epidermal support the spheroid formation serum-free culture. However, without any factors can imitate tumor environment more realistically. In this study, we aimed identify effect of deprivation on MKN-45 gastric cell line terms stemness characteristics. Materials methods: The spheroids were obtained culturing conditions, then dissociated obtain for further analyses. Self-renewal, multipotency, transformation, invasiveness, chemoresistance, expression stemness-related genes analyzed assay, soft agar colony transwell invasion chemosensitivity quantitative RT-PCR respectively. Results: Fetal bovine serum caused an increase markers OCT4, NANOG, SOX2, MUC1, CD24 CD90. Increasing functional aggressiveness-related properties, self-renewal, invasive ability, have also been observed fetal serum-growth factor-free conditions.
